What is Amprenavir

NOTE: This drug is discontinued in the US.

Amprenavir is an HIV protease inhibitor similar to indinavir and ritonavir.

Preliminary phase III data showed, in an as-treated analysis, that 88% of HIV-infected patients who received amprenavir, lamivudine, and zidovudine for 16 weeks achieved viral loads below the limit of detection (< 400 copies/mL) compared with 19% of patients who received lamivudine plus zidovudine.

Amprenavir has also been studied as part of a 4-drug regimen; mean HIV RNA plasma levels were reduced significantly from baseline after 8 weeks in HIV-infected patients treated with amprenavir, abacavir, zidovudine, and lamivudine, all administered twice daily.

Amprenavir’s twice daily administration constitutes an advantage over other protease inhibitors with more frequent administration schedules (e.g., indinavir, saquinavir).

Amprenavir was approved April 15, 1999 for use in both adults and children.

In May 2000, important labeling changes regarding the use of amprenavir oral solution were announced and included contraindications in infants and children < 4 years of age, pregnant women, patients with hepatic or renal failure, and patients taking disulfiram or metronidazole due to the high concentrations of propylene glycol present in the solution.
