Trandolapril

What is Trandolapril

Trandolapril is an oral, non-sulfhydryl-containing angiotensin converting enzyme inhibitor (ACEI). It is a prodrug for trandolaprilat which is about 8 times as potent as trandolapril in inhibiting ACE.

Trandolapril is structurally similar to enalapril and both require hydrolysis for activity. Because of the pharmacokinetics of the active metabolite, trandolaprilat, trandolapril can be administered once daily.

Trandolapril is approved for the treatment of hypertension and is under investigation for prevention of sudden death post-MI. Data from the TRACE (Trandolapril Cardiac Evaluation) study suggest trandolapril can significantly reduce sudden death post-MI, and can decrease progression to severe heart failure, however, it did not reduce the risk of recurrent myocardial infarction.

Final approval for trandolapril as a single-entity was granted May 3, 1996. A combination product containing trandolapril and verapamil SR (Tarka(R)) has also been approved for the treatment of hypertension. Trandolapril is available as Odrik in France and Gopten in Germany.
