What is Nisoldipine

Nisoldipine is a long-acting, oral, dihydropyridine calcium-channel blocker that is structurally related to nifedipine. In comparison with nifedipine, nisoldipine has greater affinity and selectivity for vascular smooth muscle, produces 5—10 times greater peripheral vasodilation, and has a longer elimination half-life.

In comparative studies of mild to moderate hypertension, nisoldipine 10 mg once daily was as effective as amlodipine 5 mg once daily.

In chronic stable angina, nisoldipine 20—40 mg once daily was as effective as amlodipine 5—10 mg once daily or immediate release diltiazem 80 mg three times daily.

Nisoldipine was initially FDA-approved for the treatment of hypertension on February 2, 1995 and was available in the US as an extended-release tablet formulation (Sular®), which allowed for once daily dosing for the treatment of hypertension or angina.

In March 2008, the traditional strengths of Sular® extended-release tablets (10, 20, 30, and 40 mg) were commercially replaced with new, lower-strength, bioequivalent formulations (8.5, 17, 25.5, 34 mg) utilizing Geomatrix™ technology.
