What is Eluxadoline
Eluxadoline is an oral mu and kappa opioid receptor agonist and a delta opioid receptor antagonist.
The drug is indicated for use in adults to treat irritable bowel syndrome with diarrhea (IBS-D).
FDA-approval was based on the results of two clinical studies involving a total of 2,426 adults with IBS-D. In these studies, patients were randomized to receive either eluxadoline 75 mg PO twice daily, 100 mg PO twice daily, or placebo.
At treatment week 12, the composite response rates were higher in patients receiving the 75 mg (24% and 29%) and 100 mg (25% and 30%) treatment dose vs. the placebo group.
Response to treatment was defined as a simultaneous improvement in daily worst abdominal pain score by at least 30% as compared to baseline weakly averages, and a reduction in the Bristol Stool Scale (BSS) to less than 5 on at least 50% of the days within a 12-week time interval.
Monitor recipients of eluxadoline for constipation; patients who develop severe constipation lasting more than 4 days should discontinue use of the drug.
