What is Danaparoid
Danaparoid is a heparinoid representing a mixture of low-molecular weight sulfated glycosaminoglycans: heparan (not heparin) sulfate (84%), dermatan sulfate (12%), and chondroitin sulfate (4%).
Danaparoid is obtained from porcine intestinal mucosa after the removal of heparin.
The mean molecular weight of danaparoid is 5500 daltons.
Only a small subfraction (4%) of heparan sulfate contains the pentasaccharide sequence common to unfractionated heparin (UFH) and low-molecular weight heparins (LMWHs).
Because danaparoid has a low rate of in vitro cross-reactivity (mean 9%) with heparin-induced antibodies, the risk of type II (immune-mediated) thrombocytopenia is much less than with UFH or LMWHs.
Danaparoid has been compared with UFH or warfarin for several clinical indications, including deep venous thrombosis (DVT) prophylaxis, DVT treatment, and heparin-induced thrombocytopenia.
It is at least as effective as warfarin for prevention of DVT in patients undergoing hip replacement surgery.
Danaparoid was approved for prophylaxis of post-operative DVT, which may lead to pulmonary embolism, in patients undergoing elective hip replacement surgery on 12/23/96.
